The protein encoded by this gene is a major apoprotein of the chylomicron. It binds to a specific liver and peripheral cell receptor, and is essential for the normal catabolism of triglyceride-rich lipoprotein constituents. This gene maps to chromosome 19 in a cluster with the related apolipoprotein C1 and C2 genes. Mutations in this gene result in familial dysbetalipoproteinemia, or type III hyperlipoproteinemia (HLP III), in which increased plasma cholesterol and triglycerides are the consequence of impaired clearance of chylomicron and VLDL remnants. Figure 1. A) Four membrane proteins were knocked out in K562 cells using XDel technology, following EditCo Bio’s proprietary workflow. Protein levels were assessed via flow cytometry at four time points: days 3, 7, 14, and 21 post-transfection, and compared to negative controls (non-edited cells or cells electroporated with Cas9 only). Membrane protein depletion remained stable over time, and cell viability was consistently high (>90%) across all samples. B) Representative flow cytometry plots showing protein levels for CD46 and CD59 in XDel knockout pools at 21 days post-editing.
